The Black Stars, who made four changes to the lineup that defeated Panama, set up in an ultra-compact 4-5-1 system that gave England little room to operate.
Benjamin Asare, starting in goal for the injured Lawrence Ati-Zigi, was untroubled for much of the contest as England laboured to break down the wall of yellow shirts.
The Black Stars' defence, marshalled by Jonas Adjetey and Jerome Opoku, stood firm against the Premier League firepower of Harry Kane, Bukayo Saka, and Jude Bellingham.
England's frustration was palpable. Despite enjoying the majority of possession, Thomas Tuchel's side managed just six shots in the first half, none of which were on target.
The Three Lions' only clear-cut chances came in the dying moments, with Asare saving well from Saka and Nico O'Reilly's header crashing against the top of the post before Kane blazed the rebound over.
The Black Stars also had their moments. Substitute Prince Adu caused panic in the England defence in the 79th minute when he broke through, with Ezri Konsa appearing to make contact inside the area. Ghana players and fans appealed loudly for a penalty, but the referee gave Konsa the benefit of the doubt.
It was a performance that embodied the spirit Carlos Queiroz had promised. The Black Stars fought together, defended as a team, and played with the discipline that the Portuguese coach had insisted would be the foundation of their World Cup campaign.
"We have 33 million lions to fight," Queiroz had said on the eve of the match. On the night, his players translated that belief into a performance that will live long in the memory.

The result leaves both teams on four points in Group L, with England retaining top spot on goal difference. Ghana will now face Croatia in their final group match on Thursday, knowing that a victory would guarantee their place in the knockout stages.
